Does Virtual Box have to be open all the time with BOINC in order to run Cosmology@home? I just started this today.

No, it doesn't have to have an active window open. Boinc will run the VM in the background. You can open the Oracle VM Virtual Box Manager to check on running virtual machine and then close that window with out harming anything.

Since I've joined Cosmology@home on May 6th, Statistics in BOINC Manager shows over 200,000 tasks performed, but I have never seen a single Transfer. Do I have the settings wrong?

I am not sure what you are talking about on transfers and tasks but, Computer 420741 seems to be running fine and returning tasks. It also has a normal amount of errors, around 1-2%. Your other listed computer also shows valid tasks.
